What causes a melanoma to develop?
  
  • Most are sporadic, not part of a specific familial pattern.
  • Solar injury is important, but not the only factor.
    • Preexisting nevus (dysplastic nevus?)
    • Heredity? No consistent genetic defect.
    • Chemical carcinogens? Rodent experiments.
  • Genetics of heritable melanoma syndromes.
    • Chromosome 1p36, CMM1 locus.
    • Chromosome 9p21, tumor supressor gene p16.
